米Oncothyreon社は3月14日、癌治療ワクチンONT-10のフェーズ1試験に初の患者登録を行ったと発表した。
同社は、2011年4月の米国癌学会（AACR）年次総会でのONT-10に関する非臨床データの発表に続き、12月には新薬治験許可申請（IND）を提出し、今回初めてヒトにおける検証が実施されることになった。
ONT-10は、肺癌、乳癌、大腸癌、前立腺癌、卵巣癌など多くの癌腫で発現する腫瘍関連抗原MUC-1を標的とするリポソーム化ワクチンだ。非臨床では、MUC-1を発現する腫瘍細胞に対して顕著な抗原抗体反応と細胞性免疫反応を示した。
また、MUC-1は同社が独自に開発したアジュバントであるPET-Lipid A（合成TLR4アゴニスト）を用いており、モノホスホリルリピドAに比べて抗原を増強する効果が高いと予測されている。
フェーズ1試験では最高48人の患者を登録し、ONT-10を8週間、毎週または隔週投与する。漸増投与により最大耐量と推奨投与量を調べた後、さらに15人追加して安全性をみる計画だ。また、試験全体を通して体液性および細胞性免疫反応も観察する。
